Update unity3d without update 2d toolkit?

Isn't possible that I update the Unity3D without update 2d toolkit plugin. Because I afraid it will affect my project.

It is, but it depends on what version of tk2d you're using. Some unity updates break some features in tk2d (and potentially your game as well) - it very much depends what version you're updating from and to, and what tk2d version you're currently on.
